EGF Receptor in Tumor Growth and Progression (Ernst Schering Foundation Symposium Proceedings #19)

The last 15 years have brought an understanding of growth and differentiation at the molecular level, expanding our knowledge of the origin and progression of cancer. Early breakthroughs defining growth control pathways came via studies of oncogenes, mutated signaling molecules that have lost the capacity to tum off their proliferative signal. Oncogenes with diverse growth-promoting activities have been discovered, covering the gamut from cell surface to nuclear signaling. Sequencing of these oncogenes revealed that they were mutated forms of captured cellular genes and displayed tyrosine kinase activity. The epidermal growth factor (EGF) receptor was the first of 40-50 transmembrane tyrosine kinase receptors to be cloned and sequenced. Beyond cell proliferation, activation of EGF receptor by its specific ligands controls important physiological processes, such as cell differentiation, apoptosis, cell migration, and cell shape. Activation of autocrine growth loops, consisting in solid human tumors of upregulated expression of EGFR together with increased production of ligands suggested its crucial role in autonomous tumor growth.

EGFR Signaling Networks in Cancer Therapy (Cancer Drug Discovery and Development)

The epidermal gro wth factor (EGF ) receptor and its downstream signal transduction networks have been implicated in the ontology and maintenance of tumor tissues, which has motivated the discovery and development of molecularly targeted anti-EGF receptor therapies. Over decades of study, the EGF receptor structure, its ligand binding domains, the physical biochemistry underlying its intrinsic tyrosine kinase catalytic function and the modular interactions with SH2, PTB, and SH3 domain containing signaling adaptor p- teins required for signal transduction, have been extensively dissected. Not only is the EGF receptor the nexus of many streams of information, but it also forms one part of a calcul- ing device by forming dimers and oligomers with the other three receptors in its family in response to at least eleven ligands (some of which are expressed in multiple forms with overlapping or quite distinct functions). This phenomenon, while recruiting to the inner surface of the cell membrane and activating multiple second messenger proteins, also allows the possibility of cross talk between these systems, permitting a further layer of information to be exchanged. Less well described are the cross re gulation of the EGF receptor and other anti-apoptotic, mitogenic and metabolic signaling systems. The study of these systems has yielded new surprises. One hurdle in these efforts has been that signal transduction pathways have frequently been defined in the generic absence of their tissue-specific or cell-interaction specific context.

Dieses Herausgeberwerk beschreibt eHealth grundsätzlich und insbesondere in Bezug auf elektronische Patientenakten, den Einsatz von mobilen medizinischen Anwendungen, Sensorik und Quantified Self bis hin zu medizinischen Geräten. Dazu liefert es einen internationalen Vergleich und gibt Hinweise, was wir in Deutschland von den Fortschritten anderer Länder lernen können. Rund 40 Studierende der DHWB Heidenheim liefern neben zwei Professoren, einer Gastprofessorin sowie Dozenten der DHBW Heidenheim und ergänzend einigen externen - auch international profilierten - Autoren einen Überblick über Trends und Entwicklungen. Sie belegen Vorteile und Zukunftsperspektiven anhand einiger Best-Practice-Beispiele, skizzieren technisch-organisatorische Voraussetzungen für Sicherheit und Privacy, diskutieren aber auch, welche Überzeugungsarbeit in gesellschaftlicher und politischer Hinsicht noch geleistet werden muss. Darüber hinaus werden zahlreiche Hinweise auf erfolgreiche Geschäftsmodellentwicklungen rund um eHealth gegeben. Kurz: eine Pflichtlektüre für alle, die die digitale Zukunft der Gesundheitsversorgung im Blick haben müssen.

eHealth: Potenziale in einem vernetzten Gesundheitsmarkt

In der Debatte um die Zukunft des Gesundheitswesens wird neuen Entwicklungen in der Informations- und Kommunikationstechnik häufig zu wenig Bedeutung beigemessen. Die anstehende Verbesserung von Prozessabläufen hin zu integrierten Versorgungsmodellen mit qualitäts- und kostenoptimierter Diagnostik, Behandlung und Nachsorge kann erheblich zur Verbesserung der Qualität medizinischer Leistungen, zur Effizienz der Versorgungs- und Verwaltungsprozesse und zur Autonomie der Patienten beitragen. Die Reform wird unterstützt durch den Aufbau einer modernen elektronischen Kommunikation zwischen den im Gesundheitswesen Tätigen (Patienten, Ärzte, Krankenhäuser, Apotheken, Versicherungsträger etc.). Der MÜNCHNER KREIS hat mit Verantwortungsträgern und Fachleuten aus Politik, Wissenschaft, Gesundheitswesen und Wirtschaft die Potenziale dieser Entwicklungen mit ihren technischen, ökonomischen und gesellschaftlichen Aspekten diskutiert und auch die Grenzen ausgelotet. This special volume contributes to the rapidly growing body of eHealth research, presenting a selection of multidisciplinary studies on the role and impacts of technology and the Internet in health communication, healthcare delivery, and patient self-management. The use of the Internet and new communication technologies have impacted nearly every aspect of life in recent years. These technologies hold tremendous promise to improve systems of healthcare and enable people to better understand their health and manage their healthcare. However, there are also risks to the use of eHealth technologies. Empirical evidence is urgently needed to examine the use and impacts of eHealth technologies and to inform targeted health communication interventions. Chapters explore both old and new challenges associated with technology-enabled care. These include the persistence of social determinants in shaping Digital Divides in access and use of eHealth technologies, the unintended consequences associated with electronic medical records and pagers on healthcare professionals’ ability to control their work time, and how self-tracking and quantification may exacerbate gendered norms of the body and health.
